The studies cover exploratory analysis of efficacy biomarkers for Aponermin based regimens and efficacy and safety evaluations of Aponermin combined with proteasome inhibitors, immunomodulatory agents, XPO1 inhibitors, and BCL-2 inhibitors, in patient populations facing substantial treatment challenges, including relapse after immunotherapy and prior multidrug exposure in relapsed/refractory multiple myeloma (RRMM).

• Aponermin is a circularly permuted variant of human TRAIL and the world’s first approved DR4/DR5 agonist. It binds death receptor 4 (DR4) and/or death receptor 5 (DR5) on the tumor cell membrane to activate the extrinsic apoptosis pathway, triggering a caspase cascade that induces p53-independent apoptosis.
• Multiple studies indicate that DR4/DR5 agonists also carry immunomodulatory activity, suppressing regulatory T cells, tumor-associated macrophages, and myeloid-derived suppressor cells within the tumor microenvironment, which may help improve the tumor immune microenvironment.
